PhD project:

Adapter proteins are a class of proteins that play a fundamental role in the context of regulating cellular homeostasis, acting as a dock for different signaling molecules, allowing them to form supramolecular complexes and become functional. In particular, the FRS2 protein (fibroblast growth factor receptor substrate 2), through its PTB domain, plays a crucial role in signaling pathways. Especially those involving the fibroblast growth factor and nerve growth factor receptor families. Overexpression of the FRS2 protein can lead to the formation of different types of tumors. Through the use of molecular biology, biochemistry and biophysics methodologies it was possible to characterize the folding mechanism of the PTB domain and the interaction between it and two ligands, FGFR1 and TRKB. It has been observed that the presence or absence of a disulfide bridge probably determines a change in the conformation of the protein. Furthermore, it has been observed that this variation does not particularly influence the interaction with physiological ligands.
