L'attività di ricerca del mio laboratorio si è anche concentrata sulla caratterizzazione delle vie di segnalazione che regolano l'apoptosi mitocondrio-dipendente e Bax-regolata in risposta ai farmaci chemioterapici e al ruolo degli oncosoppressori p53 e p73 nell'acquisizione della resistenza all'apoptosi da parte delle cellule tumorali. Questi studi hanno contribuito all'identificazione di un nuovo mutante della p53 (K351N) coinvolto nell'acquisizione della resistenza all'apoptosi in cellule di carcinoma ovarico e con difetti specifici nella tetramerizzazione, nell'attività trascrizionale e nell'esporto nucleare. Abbiamo anche contribuito alla caratterizzazione delle molecole coinvolte nell'attivazione dei fattori di trascrizione NF-kB nei linfociti T. Siamo stati tra i primi a dimostrare che la stimolazione del CD28 attiva nei linfociti T una cascata non canonica di NF-kB2 che porta al reclutamento selettivo di dimeri RelA/p52 e RelA/RelA sul promotore di diversi geni bersaglio tra cui geni di sopravvivenza e infiammatori. Questi studi hanno anche contribuito all'identificazione a livello cellulare e molecolare di diverse capacità di segnalazione tra CD28 umano e murino, sottolineando così la necessità di prestare attenzione ai dettagli molecolari quando si trasferiscono i risultati da modelli murini all’uomo.
Più recentemente ci siamo concentrati sul ruolo del CD28 nella regolazione dei processi metabolici e infiammatori nella Sclerosi Multipla (SM), evidenziando un importante contributo della PI3K di classe 1A associata al CD28 nella riprogrammazione dei processi metabolici che mantengono/amplificano il fenotipo infiammatorio dei linfociti T periferici e suggeriscono la PI3K di classe 1A un valido bersaglio terapeutico per la SM.
La nostra ricerca corrente è finalizzata alla caratterizzazione del ruolo del CD28 nella riposta infiammatoria sistemica attivata dai superantigeni batterici dello Staphyloccoccus aureus, evidenziando il suo ruolo del CD28 e del TCR nella produzione da parte dei linfociti T di citochine infiammatorie in risposta al SEB e SEA
