Titolo della tesi: Role of miR-146a in the inflammatory response of macrophages from healthy and cystic fibrosis individuals

Macrophages are tissue resident cells that mediate the first response to infection through the elimination of foreign materials and the orchestration of the inflammatory response. MicroRNAs have emerged as key regulators of several aspects of macrophage biology and their deregulation has been associated to the pathogenesis of several human inflammatory diseases. Cystic fibrosis (CF) is an autosomal recessive genetic disease, caused by mutations in the CF transmembrane conductance regulator gene (CFTR), characterized by an increased susceptibility to bacterial lung infections and exaggerated inflammatory responses. Defective bactericidal activity and exaggerated inflammatory responses have been attributed to CF macrophages. To verify whether miRNA dysregulation underlies the functional abnormalities of CF macrophages we performed miRNA profiling in such cells and identified 22 differentially expressed miRNAs. Among these, we analysed the consequences of increased miR-146a expression on the inflammatory response of CF macrophages demonstrating that miR-146a overexpression in CF is functional to restrict inflammation. In addition, to elucidate the role of miR-146a in the inflammatory response of human macrophages we evaluated the cytokine response and the modulation of TLR4 signalling molecules in LPS stimulated macrophages following inhibition of endogenous miR-146a. Results demonstrate that miR-146a mainly modulates TNF-a secretion and TRAF6 protein expression. I macrofagi sono cellule residenti nei tessuti, che mediano la prima risposta alle infezioni mediante l’eliminazione delle sostanze estranee e l’attivazione della risposta infiammatoria. I microRNA regolano diversi aspetti della biologia dei macrofagi e la loro disregolazione è stata associata alla patogenesi di diverse malattie infiammatorie umane. La fibrosi cistica (FC) è una malattia genetica autosomica recessiva, causata da mutazioni nel canale regolatore della conduttanza trans-membrana (CFTR) e caratterizzata da una aumentata suscettibilità ad infezioni batteriche polmonari e da una esagerata risposta infiammatoria. Difetti nella capacità battericida e della risposta infiammatoria sono stati attribuiti ai macrofagi FC. Per verificare se la disregolazione dei miRNAs fosse alla base delle alterazioni dei macrofagi FC è stato caratterizzato il profilo di espressione dei miRNA in tali cellule identificando 22 miRNA differenzialmente espressi. Fra questi, abbiamo studiato le conseguenze dell’aumentata espressione del miR-146a sulla risposta infiammatoria dei macrofagi FC dimostrando che questo contribuisce al contenimento dell’infiammazione. Inoltre, per definire il ruolo del miR-146a nella risposta infiammatoria dei macrofagi umani abbiamo analizzato la produzione di citochine e la modulazione di molecole di segnalazione del TLR4 nei macrofagi stimolati con LPS e in cui era stato inibito il miR-146a endogeno. I dati ottenuti dimostrano che il miR-146a modula principalmente il TNF-α e l’espressione della proteina TRAF6.
