Ricerca: Gli effetti degli inibitori di TPC2 sulla via dell'autofagia durante l'osteoblastogenesi e l'osteoclastogenesi

L'autofagia è coinvolta nell'osteogenesi e il suo disturbo è associato all'osteoporosi. Durante la differenziazione degli osteoclasti, l'espressione dei canali a due pori di tipo 2 (TPC2) aumenta e ci sono alcune prove che il silenziamento di TPC2 possa inibire l'espressione dei marcatori di autofagia. Anche un'altra famiglia di proteine chiamata galectine è coinvolta nell'osteogenesi e alcuni studi hanno riscontrato la cooperazione tra ULK1 e galectine durante la differenziazione delle cellule ossee. Sulla base di queste informazioni, gli obiettivi di questo studio sono: 1. Valutare gli effetti degli inibitori di TPC2 sulla via dell'autofagia nell'osteogenesi, 2. Comprendere la correlazione tra galectina e TPC2 durante l'osteogenesi, 3. Studio dei parametri ossei, TPC2 e l'espressione della galectina in topi beclin1 knockout.
