Research activities
Research sectors
The mechanism of action of proteins was investigated by using protein crystallography and biochemical characterization. This included the structure-function relationship of enzymes and transporters as well as their role within signalling networks that involves protein-protein recognition and complex formation.
The knowledge of the molecular mechanism exploited by proteins to carry out their function and involved in pathological processes is the starting point to design novel, targeted therapeutic strategies. In this contest, the main endeavour has been the determination of protein structures by protein crystallography and, more recently by single particle cryo-electron microscopy. Protein biochemical and biophysical characterization was carried out using a wide array of methods (kinetics, protein engineering, spectroscopy). Moreover, Andrea Ilari’s research have been focused on structure-based drug design targeting proteins involved in different pathology. The main topics of Andrea Ilari’s research have been:
1) Structrure-function relationships in E. coli flavohemoglobin and in the following truncated hemoglobins: Bacillus subtilis truncated hemoglobin and Thermobifida fusca truncated hemoglobin.
2) The structure function relationships in Sorcin (SOluble Resistance related Calcium bInding proteiN) a protein involved in excitation-contraction-relaxation processes in the heart through the interaction with several Ca2+ channels and involved in MuntiDrug Resistance in cancer.
3) Structure function relationships in Dps (DNA binding proteins from starved cells) cage-like proteins involved in the bacterial-DNA protection from oxidative damage and used as bioreactor to produce metal-based nanoparticles.
4) Structure-function relationship in proteins involved in the trypanothione metabolism, a molecule which protect the Leishmania parasites (responsible for Leishmaniasis) from the ROS (Reactive Oxygen Species) produced by macrophages during the infection .
5) Structure-based drug design targeting Trypanothione Reductase a protein involved in trypanothione metabolism and essential for the Leshmania parasite survival in the human host. The design of new lead compound isperformed using modern crystallographic techniques as Fragment Based Screening.
6) Study of the proteins involved in Zinc transport in pathogenic bacteria. The study aims at designing a new antibiotic against Salmonella enterica which, as all the gram negative bacteria, develops very easily drug resistance.
7) Structure-function relationship in Sigma1 R (S1R) a neuroprotective protein. The study aims at finding new S1R agonist to be used to project new drugs to cure neurodegenerative diseases.
